In this report, the site-specific co-delivery of green tea/aluminium magnesium silicate (AMS) was reported and the specific target delivery was achieved orally. The new co-precipitation process was developed to synthesis the green tea/AMS hybrid complex and using energy-dispersive X-ray spectroscopy, Fourier-transform infrared spectroscopy and Raman confirmed its successful synthesis. The blood biocompatibility of the green tea/AMS was tested using chicken blood, and the compound is safe up to 500 mg/ml. After mixed with hydroxypropyl methylcellulose phthalate, the oral beads were synthesised using a linking agent. The oral beads underwent different pH-based dissolution studies and the results indicated that the beads specifically dissolved in gastric pH (6.5). The pharmaco kinetic studies were performed to estimate the delivery kinetics. The results revealed that the beads underwent as per the Higuchi model. The anticoccidial effects of the beads were tested using chicken. The animal studies were performed in two different modes such as prophylactic treatment and active treatment after Eimeria species challenge. The results indicated that the prophylactic treatment with beads 100% protected the chicken and the active treatment with beads after the Eimeria challenge significantly protected against the intestinal damage and it also enhanced the anticoccidial effect.

Sulphonylureas are extensively used in the treatment of type II diabetes; however, these drugs have complications of hypoglycaemia and weight gain. The current study aims at developing a potent antidiabetic drug that has lesser side effects and better management of its associated conditions. Zinc oxide nanoparticles (ZnO NPs) were synthesised using Syzygium cumini seed extract with an average size of 18.92 nm. In vitro studies on rat insulinoma (RIN-5F) cells revealed that cells treated with synthesised ZnO NPs showed a dose-dependent increase in insulin secretion. Streptozotocin-fructose-induced type II diabetic rats treated with ZnO NPS exhibited a significant reduction (p < 0.01) in the blood glucose levels, total cholesterol, triglycerides, and low-density lipoprotein levels and increase (p < 0.01) in serum insulin and liver antioxidant enzyme levels proclaiming its role as a hypoglycaemic and hypolipidaemic drug. Treatment of ZnO NPs in diabetic rats exhibited an increased number of beta cells which was responsible for its increased insulin levels and reduced glucose levels. From the overall observations, biosynthesised ZnO NPs exhibited an efficacious hypoglycaemic effect in diabetic rats, so it can be suggested as a potent antidiabetic drug.

In the modern pace of the world, food safety is a major concern. In this work, a simple chemiresistive type gas sensor was fabricated to detect Escherichia Coli (E. coli) bacteria. Polyaniline (PANI) films were deposited on the indium tin oxide substrate by an electrochemical deposition method. TiO₂ nanoparticles were synthesised by facile hydrothermal method. PANI films were modified using hydrothermally prepared TiO₂ nanoparticles by a spin coating method. X-ray diffraction (XRD), field emission scanning electron microscope (FESEM), Fourier transform infrared (FTIR) and ultraviolet–visible spectrophotometer techniques were used to characterise the PANI/TiO₂ nanocomposites. The peaks obtained in the XRD pattern confirmed the anatase phase of TiO₂ nanoparticles. FESEM analysis showed the nanofibrous structure of the nanocomposite. The FTIR characteristic peaks confirmed the formation of the nanocomposite. The electrical resistance of the sensors was evaluated as a function of the bacterial concentration. The PT2 (TiO₂ coated 5 times on PANI) in comparison with PT1 (TiO₂ coated 3 times on PANI) exhibited good sensitivity to the gas molecules at room temperature. The p-n junction at PANI/TiO₂ interface improved the physical adsorption of gas molecules. Since no specific antibodies or receptors are used, the sensor has the potential for adaptation to real-life applications. Thus low cost, real-time, portable, reusable and sensitive bacteria sensors were fabricated and tested.

In this study, probiotic yeast Saccharomycopsis fibuligera (S. fibuligera) VIT-MN04 was encapsulated with wheat bran fibre (WBF) and exopolysaccharide (EPS) along with 5% polyvinylpyrrolidone (PVP) using electrospinning technique for easy gastrointestinal transit (GIT). The electrospinning materials viz. WBF (10%), EPS (15%), PVP (5%) and electrospinning parameters viz. applied voltage (10 kV) and tip to collector distance (15 cm) were optimised using response surface methodology to produce fine nanofibres to achieve maximum encapsulation efficiency (100%) and GIT tolerance (97%). The probiotic yeast was successfully encapsulated in nanofibre and investigated for potential properties. The survival of encapsulated S. fibuligera VIT-MN04 was increased compared to the free cells during in vitro digestion. In addition, encapsulated yeast cells retained their viability during storage at 4°C for 56 days. The nanofibres were characterised using scanning electron microscopy, atomic force microscopy, X-ray diffraction, thermogravimetric analysis, zeta potential analysis and Fourier transform infrared spectroscopy followed by gas chromatography–mass spectrometry and nuclear magnetic resonance analysis. This work provides an efficient approach for encapsulation of probiotic yeast with the nanofibres which can also broaden the application of the prebiotic like WBF providing an idea for the efficient preparation of functional synbiotic supplements in the food industry.

Candida albicans (C. albicans) infection shows a growing burden on human health, and it has become challenging to search for treatment. Therefore, this work focused on the antifungal activity, and cytotoxic effect of biosynthesised nanostructures on human ovarian tetracarcinoma cells PA1 and their corresponding mechanism of cell death. Herein, the authors fabricated advanced biosynthesis of uncoated α-Fe₂O₃ and coated α-Fe₂O₃ nanostructures by using the carbohydrate of Spirulina platensis. The physicochemical features of nanostructures were characterised by UV–visible, high resolution transmission electron microscopy, Fourier transform infrared spectroscopy, and X-ray diffraction. The antifungal activity of these nanostructures against C. albicans was studied by the broth dilution method, and examined by 2′, 7′-dichlorofluorescein diacetate staining. However, their cytotoxic effects against PA1 cell lines were evaluated by MTT and comet assays. Results indicated characteristic rod-shaped nanostructures, and increasing the average size of α-Fe₂O₃@ZnO nanocomposite (105.2 nm × 29.1 nm) to five times as compared to α-Fe₂O₃ nanoparticles (20.73nm × 5.25 nm). The surface coating of α-Fe₂O₃ by ZnO has increased its antifungal efficiency against C. albicans. Moreover, the MTT results revealed that α-Fe₂O₃@ZnO nanocomposite reduces PA1 cell proliferation due to DNA fragmentation (IC₅₀ 18.5 μg/ml). Continual advances of green nanotechnology and promising findings of this study are in favour of using the construction of rod-shaped nanostructures for therapeutic applications.

In the present study, biogenic silica nanoparticles (bSNPs) were synthesized from groundnut shells, and thoroughly characterized to understand its phase, and microstructure properties. The biopolymer was synthesized from yeast Wickerhamomyces anomalus and identified as Poly (3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV) by GC-MS and NMR analysis. The bSNPs were reinforced to fabricate PHBV/SiO₂ nanocomposites via solution casting technique. The fabricated PHBV/SiO₂ nanocomposites revealed intercalated hybrid interaction between the bSNPs and PHBV matrix through XRD analysis. PHBV/SiO₂ nanocomposites showed significant improvement in physical, chemical, thermo-mechanical and biodegradation properties as compared to the bare PHBV. The cell viability study revealed excellent biocompatibility against L929 mouse fibroblast cells. The antibacterial activity of PHBV/SiO₂ nanocomposites was found to be progressively improved upon increasing bSNPs concentration against E. coli and S. aureus.

Honokiol (HK) is a natural product isolated from the bark, cones, seeds and leaves of plants belonging to the genus Magnolia. It possesses anti-cancer activity which can efficiently impede the growth and bring about apoptosis of a diversity of cancer cells. The major concerns of using HK are its poor solubility and lack of targeted drug delivery. In this study, a combinatorial drug is prepared by combining HK and camptothecin (CPT). Both CPT and HK belong to the Magnolian genus and induce apoptosis by cell cycle arrest at the S-phase and G1 phase, respectively. The combinatorial drug thus synthesised was loaded onto a chitosan functionalised graphene oxide nanoparticles, predecorated with folic acid for site-specific drug delivery. The CPT drug-loaded nanocarrier was characterised by X-ray diffractometer, scanning electron microscope, transmission electron microscope, UV–vis spectroscopy and fluorescence spectroscopy, atomic force microscopy. The antioxidant properties, haemolytic activity and anti-inflammatory activities were analysed. The cellular toxicity was analysed by 3-(4,5-Dimethylthiazol-2-yl)-2,5-Diphenyltetrazolium Bromide (MTT assay) and Sulforhodamine B (SRB) assay against breast cancer (MCF-7) cell lines.

The therapeutics for type 2 diabetes mellitus has emerged in the current century towards nanomedicine incorporated with plant active compounds. In this study, Tinospora cordifolia loaded poly (D, L-lactide) (PLA) nanoparticles (NPs) were evaluated in vivo for their anti-hyperglycemic potency towards streptozotocin-induced type 2 diabetic rats. T. cordifolia loaded PLA NPs were synthesised by the double solvent evaporation method using PLA polymer. The NPs were then characterised and administrated orally for 28 successive days to streptozotocin-induced diabetic rats. The PLA NPs had significant anti-diabetic effects which were equal to the existing anti-diabetic drug glibenclamide. The antidiabetic activity is due to the synergism of compounds present in stem extract of the plant which reduced the side effects and anti-diabetic.

In the present work cassava starch/agar Ag and ZnO nanocomposite films were prepared by the solution casting method. The structural, physical and antimicrobial properties of the nanocomposite films were studied as a function of the concentration of Ag and ZnO nanoparticles. The results of the thermogravimetric analysis showed 8–15% degradation of both the nanocomposite films at 150°C endorsing the thermal stability of the films. Scanning electron microscopic analysis reveals the uniform blending of Ag and ZnO nanoparticles with a starch/agar matrix with tiny waves like appearance on the surface. The incorporation of Ag and ZnO nanoparticles in the film was found to reduce the moisture content, water solubility and water vapour permeability with increase in the concentration of Ag and ZnO nanoparticles. The growth kinetics study of Pseudomonas aeruginosa and Staphylococcus aureus in the presence of Ag and ZnO blended nanocomposite films showed promising results especially against Gram-negative P. aeruginosa. Thus, the film synthesised in the present study bears the potential to be used as active packaging material to prevent food from bacterial contamination and spoilage.

The authors report a novel, effective and enhanced method of conjugating anticancer drug, paclitaxel and gallic acid with magnetosomes. Here, anticancer drugs were functionalised with magnetosomes membrane by direct and indirect (via crosslinkers: glutaraldehyde and 3-aminopropyltriethoxysilane) adsorption methods. The prepared magnetosome–drug conjugates were characterised by Fourier transform infrared, zeta potential, field-emission scanning electron microscope and thermogravimetric analysis/differential scanning calorimetry. The drug-loading efficiency and capacity were found to be 87.874% for paclitaxel (MP) and 71.3% for gallic acid (MG), respectively as calculated by ultraviolet spectroscopy and high-performance liquid chromatography. The drug release demonstrated by the diffusion method in phosphate buffer (PBS), showing a prolonged drug release for MP and MG, respectively. The cytotoxicity effect of the MP and MG displayed cytotoxicity of 69.71%, 55.194% against HeLa and MCF-7 cell lines, respectively. The reactive oxygen species, acridine orange and ethidium bromide and 4, 6-diamidino-2-phenylindole staining of the drug conjugates revealed the apoptotic effect of MP and MG. Further, the regulation of tumour suppressor protein, p53 was determined by western blotting which showed an upregulation of p53. Comparatively, the magnetosome–drug conjugates prepared by direct adsorption achieved the best effects on the drug-loading efficiency and the increased percentage of cancer cell mortality and the upregulation of P53. The proposed research ascertains that magnetosomes could be used as effective nanocarriers in cancer therapy.

Preparation of manganese ferrite (MnFe₂O₄) nanorods by the reduction of akaganeite seeds in the presence of oleylamine is reported. The Mn-doped β-FeOOH akaganeite seeds have been processed by the hydrolysis of metal-chloride salts in the presence of polyethylenimine (PEI) surfactant. The hydrophobic oleylamine capped nanorods are made hydrophilic using trisodium citrate as a phase transferring agent. The nanorods form with an aspect ratio of 5.47 and possess a high magnetisation value of 69 emu/g at an applied magnetic field of 1.5 T. The colloidal water dispersion of nanorods exhibits superior heating efficiency by the application of alternating magnetic field (AMF). A specific absorption rate value of 798 W/g is achieved at an applied AMF of field strength 500 Oe and frequency 316 kHz. Further, the citrate functionalised nanorods are capable of attaching with doxorubicin (DOX) electrostatically with a loading efficiency of 97% and the drug release is pH responsive. The DOX loaded nanorods show a promising effect on the apoptosis of MCF-7 as experimented in vitro.

Scaffolds based on chitosan (CTS), collagen (Coll) and glycosaminoglycans (GAG) mixtures cross-linked by tannic acid (TA) with bioglass 45S5 addition were obtained with the use of the freeze-drying method. The prepared scaffolds were characterised for morphology, mechanical strength and degradation rate. Moreover, cell viability on the obtained scaffolds was measured with and without the presence of ascorbic acid and dexamethasone. The main purpose of the research was to compare the effectiveness of bioglass 45S5 influence on the physicochemical and biological properties of scaffolds. The results demonstrated that the scaffolds based on the blends of biopolymers cross-linked by TA are stable in an aqueous environment. Scanning electron microscope images allowed the observation of a porous scaffold structure with interconnected pores. The addition of bioglass nanoparticles improved the mechanical properties and decreased the degradation rate of composite materials. The biological properties were improved for 20% tannic acid addition compared to 5%. However, the addition of bioglass 45S5 did not change to cells response significantly.

The present investigation aims for the synthesis of copper oxide nanoparticles (CuO NPs) using Nilgirianthus ciliatus plant extract. The obtained CuO NPs were characterised by X-ray diffraction, Fourier transform infrared spectrum, ultraviolet–visible spectroscopy, photoluminescence, scanning electron microscopy and transmission electron microscopy analysis. Significant bacterial activity was manifested by CuO nanoparticles against both Gram-positive (Staphylococcus aureus and Staphylococcus mutans) and Gram-negative (Escherichia coli and Pseudomonas aeruginosa) bacteria. The synthesised CuO NPs have good cytotoxicity against both human breast cancer cell line (MCF-7) and lung cancer cell line (A549) with minimum cytotoxic effect on normal L929 (fibroblast) cell lines.

Listeriosis through contaminated food is one of the leading causes of premature deaths in pregnant women and new born babies. Here, the authors have developed a magnetosomes-based biosensor for the rapid, sensitive, specific and cost-effective detection of Listeria monocytogenes from food sample. Magnetosomes were extracted from Magnetospirillum sp. RJS1 and then directly bound to anti-Listeriolysin antibody (0.25–1 µg/ml), confirmed in spectroscopy. Listeriolysin (LLO) protein (0.01–7 µg/ml) was optimised in enzyme-linked immunosorbent assay. Magnetosomes was conjugated with LLO antibody (0.25 µg/ml) in optimum concentration to detect LLO protein (0.01 µg/ml). Magnetosomes–LLO antibody complex was 25% cost effective. The magnetosomes–LLO antibody complex was directly stabilised on screen printed electrode using external magnet. The significant increase in resistance (R _CT value) on the electrode surface with increase in concentration of LLO protein was confirmed in impedance spectroscopy. The L. monocytogenes contaminated milk and water sample were processed and extracted LLO protein was detected in the biosensor. The specificity of the biosensor was confirmed in cross-reactivity assay with other food pathogens. The detection limit of 10¹ Cfu/ml in both water and milk sample manifests the sensitive nature of the biosensor. The capture efficiency and field emission scanning electron microscopy confirmed positive interaction of Listeria cells with magnetosomes–antibody complex.

Metal nanoparticles have generated great interest due to their excellent optical and chemical properties. The widely used chemical method for synthesising nanoparticles involves capping agents for colloidal stability. However, there are scarce reports on the application of metal nanoparticles synthesised without using capping agents. Hence, there is a need to develop pristine nanoparticles devoid of capping that can be used for translational research. Here, the authors developed a facile and rapid method for synthesising bare metal nanoparticles (platinum/silver/gold) that are chemically reactive and stable for a month upon storage. They synthesised bare metal nanoparticles of sub-15 nm and characterised using standard techniques (UV–VIS-NIR/DLS/zeta//TEM/XRD). They assessed the safety of the synthesised nanoparticles on the liver carcinoma cell line (HepG2). Bare gold and platinum nanoparticles were non-toxic in comparison to bare silver nanoparticles. Bare metal nanoparticles were also checked for metal detection wherein antimony, mercury and chromium were detected using bare gold and silver nanoparticles. The spectroscopic shifts of the nanoparticles when bound to metals resulted in blue and red shifting of the plasmon band, indicating the sensing of metals. These results show that bare metal nanoparticles have the potential to emerge as a promising candidate for biomedical and sensing applications.

In the present investigation, Rheum emodi roots extract mediated magnesium hydroxide nanoparticles [Mg(OH)₂ NPs] through the bio-inspired experimental technique were synthesised. Mg(OH)₂ NPs were characterised by using various characterisation techniques such as field emission scanning electron microscopy, transmission electron microscopy (TEM), Fourier transform infrared spectroscopy (FTIR), and ultraviolet–visible spectroscopy. The formation of Mg(OH)₂ NPs was confirmed by X-ray diffraction. The structural analysis confirmed the hexagonal crystal symmetry of Mg(OH)₂ NPs with space group P-3m1 and space group no. 164 using the Rietveld refinement technique. TEM micrographs illustrated the nano-size formation of Mg(OH)₂ NPs of spherical shape and size ∼14.86 nm. With the aid of FTIR data, plant metabolites such as anthraquinones have been identified as a stabilising and reducing agent for the synthesis of biogenic Mg(OH)₂ NPs. The synthesised Mg(OH)₂ NPs showed antimicrobial and cytotoxic potential against Gram-negative and Gram-positive bacteria such as Escherichia coli (ATCC 25922) and Staphylococcus aureus (ATCC 25923) and MDA-MB-231 human breast cancer cell lines.

The mechanism of nanoparticles (NPs) synthesis is a crucial prerequisite for the engineering of desirable material properties and is elusive due to the difficulty of studying the synthesis path and also due to lack of published work. The present study moves beyond the current conventional study of characterisation of NPs synthesis. It focuses on study of classical kinetics of metal NP using silver as a model metal to perform the required experiments, it aimed at understanding the crystallisation process which is the major root for the synthesis of metal NP. The authors have chosen biological approach to explain the process of synthesis of metal NP using inductively coupled plasma – optical emission spectroscopy that directly analyses the concentration of metal NPs. Alpha-amylase solution, when incubated with silver nitrate solution, turned brown at a specific concentration of enzyme and substrate, which shows the formation of silver NPs. Inductively coupled plasma data and dynamic light scattering spectra were studied to understand the kinetics and the kinetics model was obtained. The enthalpy (ΔH*), activation energy (ΔE*), and equilibrium constant (K) were calculated for understanding the thermodynamics of the reaction. The process of NP synthesis is dependent on the kinetics of the reaction, and other process parameters limit the thermodynamics of the process.

Umbilical cord-derived mesenchymal stem cells (UCDMSC) are attractive candidates for cell-based regenerative medicine. However, they are susceptible to replicative senescence during repetitive passaging for in-vitro expansion and induced senescence in an oxidative, inflammatory microenvironment in vivo. Aim of this study is to investigate if honey-incorporated matrices can be employed to reduce senescence of UCDMSC. Matrices were prepared by electrospinning solutions of honey with poly-vinyl alcohol (PVA). PVA:honey matrices exhibited free radical scavenging activity. Culture of UCDMSC on PVA:honey matrices showed improvement in cell proliferation compared to pure PVA nanofibres. Expression of vimentin indicated that mesenchymal phenotype is preserved after culturing on these matrices. Further, UCDMSC were serially subcultured and cells of two passages (P2 and P6) were evaluated for reactive oxygen species (ROS) load and senescence parameters. P6 cells showed a higher ROS load and β-galactosidase (β-gal) positive senescent cells compared to P2. However, culturing on PVA:honey substrates significantly reduced both ROS and β-gal markers compared to cells on PVA substrates. Honey contains several antioxidant and anti-inflammatory components, which can reduce the ROS-related senescence. Thus, it is concluded that honey containing nanofibres can be effective substrates for stem cell-based wound healing and regenerative medicine.

Effective and targeted delivery of the antitumour drugs towards the specific cancer spot is the major motive of drug delivery. In this direction, suitably functionalised magnetic iron oxide nanoparticles (NPs) have been utilised as a theranostic agent for imaging, hyperthermia and drug delivery applications. Herein, the authors reported the preparation of multifunctional polyethyleneglycol-diamine functionalised mesoporous superparamagnetic iron oxide NPs (SPION) prepared by a facile solvothermal method for biomedical applications. To endow targeting ability towards tumour site, folic acid (FA) is attached to the amine groups which are present on the NPs surface by 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide hydrochloride/N-hydroxysuccinimide chemistry. FA attached SPION shows good colloidal stability and possesses high drug-loading efficiency of ∼ 96% owing to its mesoporous nature and the electrostatic attachment of daunosamine (NH₃ ⁺) group of doxorubicin (DOX) towards the negative surface charge of carboxyl and hydroxyl group. The NPs possess superior magnetic properties in result endowed with high hyperthermic ability under alternating magnetic field reaching the hyperthermic temperature of 43°C within 223 s at NP's concentration of 1 mg/ml. The functionalised NPs possess non-appreciable toxicity in breast cancer cells (MCF-7) which is triggered under DOX-loaded SPION.

Green synthesis of organic Pt-nanocomposite was accomplished using carboplatin as a precursor and novel biopolymer – gum kondagogu (GK) as a reducing agent. The synthesised GK stabilised organic Pt-nanocomposite (GKCPt NC) was characterised by different analytical techniques such as ultraviolet–visible spectroscopy, nanoparticle analyser, scanning electron microscopy and energy dispersive X-ray analysis, X-ray diffraction (XRD), Fourier-transform infrared spectroscopy, transmission electron microscopy (TEM), X-ray photoelectron spectroscopy (XPS) and inductively coupled plasma optical emission spectrophotometer. The XRD pattern established the amorphous nature of GKCPt NC. TEM analysis revealed the homogeneous, monodisperse and spherical nature, with Pt metal size of 3.08 ± 0.62 nm. The binding energy at 71.2 and 74.6 eV show the presence of metallic platinum, Pt(0) confirmed by XPS studies. Further, in vitro radical scavenging and antitumour activity of GKCPt NC have been investigated. In comparison to GK and carboplatin, GKCPt NC showed superior 1, 1-diphenyl-2-picrylhydrazyle activity of 87.82%, whereas 2, 2-azinobis-(3-ethylbenzthinzoline-6-sulphonic acid) activity was 38.50%, respectively. In vitro studies of the antitumour property of GK, GKCPt NC and carboplatin were evaluated by potato disc tumour bioassay model. The efficacy of synthesised GKCPt NC concentration (IC₅₀) on tumour inhibition was found to be 2.04-fold lower as compared to carboplatin. Overall, the synthesised GKCPt NC shows both antitumour and antioxidant properties when compared to the original drug – carboplatin and might have promising applications in cancer therapy.

Tissue plasminogen activator (tPA) a thrombolytic agent is commonly used for digesting the blood clot. tPA half-life is low (4–6 min) and its administration needs a prolonged continuous infusion. Improving tPA half-life could reduce enzyme dosage and enhance patient compliance. Nano-carries could be used as delivery systems for the protection of enzymes physically, enhancing half-life and increasing the stability of them. In this study, chitosan (CS) and polyethylene glycol (PEG) were used for the preparation of CS-g-PEG/tPA nanoparticles (NPs) via the ion gelation method. Particles’ size and loading capacity were optimised by central composite design. Then, NPs cytotoxicity, release profile, enzyme activity and in vivo half-life and coagulation time were investigated. The results showed that NPs does not have significant cytotoxicity. Release study revealed that a burst effect happened in the first 5 min and resulted in releasing 30% of tPA. Loading tPA in NPs could decrease 25% of its activity but the half-life of it increases in comparison to free tPA in vivo. Also, blood coagulation time has significantly affected (p-value = 0.041) by encapsulated tPA in comparison to free tPA. So, CS-g-PEG/tPA could increase enzyme half-life during the time and could be used as a non-toxic candidate delivery system for tPA.
